Why Antidepressants Stop Working and What to Do Next

You found a medication that helped. Maybe it took a few tries to get there, but eventually something worked. There is no single explanation that applies to every patient, but the most well-supported mechanisms include the following. Neurobiological adaptation. The brain is constantly adjusting to its chemical environment. When an antidepressant increases serotonin availability in the synaptic cleft, the brain responds over time by downregulating serotonin receptors, reducing their number or sensitivity to compensate for the increased stimulation.
